چکیده

The ovarian follicular fluid was found to contain steroidogenesis stimulatory protein similar to albumin from human and buffalo. Therefore, the albumins from various species, commercial and purified, were studied for their steroidogenic effect on progesterone secretion by granulosa cells from buffalo ovaries, during culture. A dose of 20 μg of bovine serum albumin was optimum to exhibit maximum progesterone secretion on day 6 of culture, in medium (350 μl) containing 10 cells. Among commercial albumins, chicken albumin showed highest effect on progesterone secretion, which was followed by albumins from goat, bovine, human, sheep and rat, respectively at day 6 of culture. The albumins were also purified from blood serum of buffalo, goat and rat using salt fractionation, ion-exchange chromatography, gel filtration and SDS-PAGE. The highest stimulatory effect on progesterone secretion was shown by albumin purified from buffalo blood serum and lowest by that from rat blood. Comparatively the buffalo and goat albumins were more biologically active than commercial albumins. The presence of some active molecules conjugated with freshly purified albumins may be responsible for better stimulatory effect. (Asian-Aust. J. Anim. Sci. 2002. Vol 15, No. 11 : 1559-1563)

Effect of Follicular Fluid Proteins and Gonadotropins on Progesterone Secretion by Buffalo Granulosa Cells In vitro

In the mammalian ovary the follicular fluid contains proteins and peptides which play an important role in growth, development and maturation of oocytes. The gonadotropins and some other factors work synergistically and regulate ovarian functions. In the present study the effect of follicular fluid proteins (FFP) and gonadotropins on progesterone secretion by granulosa cells (GC) from buffalo o...

Effects of cholesterol, FSH and LH on steroidogenic activity of cat granulosa cells cultured in vitro

The aim of this study was to examine the effects of 22R-hydroxycholesterol (22R-HC), folliclestimulating hormone (FSH) and luteinizing hormone (LH) on estradiol and progesterone production by cat granulosa cells. Granulosa cells from follicles were collected and cultured for up to 5 days in 24 well plates containing Dulbecco’s Modified Eagle’s Medium (DMEM)/HAM F-12 supplemented with 10 M andro...

Follicle-stimulating hormone and luteinizing hormone/chorionic gonadotropin stimulation of vascular endothelial growth factor production by macaque granulosa cells from pre- and periovulatory follicles.

Granulosa cells in the ovulatory follicle express messenger ribonucleic acid encoding vascular endothelial growth factor (VEGF), an agent that may mediate the neovascularization of the developing corpus luteum, but it is not known whether luteinizing granulosa cells synthesize and secrete VEGF during the periovulatory interval. Studies were designed to evaluate the effects of an in vivo gonadot...

نقش ارتباطات بین سلولی در القای گیرنده LH در موش رات

The cyclical changes in the gonadotropins stimulate ovarian follicular development either to the ovulatory stage or to undergo atresia. One such intrafollicular factor may be inter-cellular communication via gap junctions. We have examined the effects of two agents (retinoids and alkanols), known to disrupt or uncouple gap junction, on FSH-stimulated LH receptor induction and progestrone synthe...
